Amoeba may offer key clue to photosynthetic evolution

ScienceDaily (Feb. 27, 2012) ? The major difference between plant and animal cells is the photosynthetic process, which converts light energy into chemical energy. When light isn't available, energy is generated by breaking down carbohydrates and sugars, just as it is in animal and some bacterial cells. Two cellular organelles are responsible for these two processes: the chloroplasts for photosynthesis and the mitochondria for sugar breakdown. New research from Carnegie's Eva Nowack and Arthur Grossman has opened a window into the early stages of chloroplast evolution.

Their work is published online by the Proceedings of the National Academy of Sciences in the week of February 27-March 2.

It is widely accepted that chloroplasts originated from photosynthetic, single-celled bacteria called cyanobacteria, which were engulfed by a more complex, non-photosynthetic cell more than 1.5 billion years ago. While the relationship between the two organisms was originally symbiotic, over evolutionary time the cyanobacterium transferred most of its genetic information to the nucleus of the host organism, transforming the original cyanobacterium into a chloroplast that is no longer able to survive without its host. A similar process resulted in the creation of mitochondria.

To sustain the function of the organelle, proteins encoded by the transferred genes are synthesized in the cytoplasm, or cell's interior, and then imported back into the organelle. In most systems that have been studied, the transport of proteins into the chloroplast occurs through a multi-protein import complex that enables the proteins to pass through the envelope membranes that surround the chloroplast.

Clearly the events that gave rise to chloroplasts and mitochondria changed the world forever. But it is difficult to research the process by which this happened because it took place so long ago. One strategy used to elucidate the way in which this process evolved has relied on identifying organisms for which the events that resulted in the conversion of a bacterium into a host-dependent organelle occurred more recently.

Nowack and Grossman focused their research on a type of amoeba called Paulinella chromatophora, which contains two photosynthetic compartments that also originated from an endosymbiotic cyanobacterium, but that represent an earlier stage in the formation of a fully evolved organelle.

These compartments, called chromatophores, transferred more than 30 of the original cyanobacterial genes to the nucleus of the host organism. While gene transfer has been observed for other bacterial endosymbionts, the function of the transferred genes has been unclear, since it does not appear that the endosymbionts (in contrast to organelles) are equipped to recapture those proteins, because they do not have appropriate protein import machineries.

The Carnegie team honed in on three of the P. chromatophora transferred genes, which encode proteins involved in photosynthesis, a process localized to the chromatophore. They set out to determine whether these proteins are synthesized in the cytoplasm of the amoeba and whether the mature proteins became localized to the chromatophore.

Using an advanced array of research techniques, they were able to determine that these three proteins are synthesized in the cytoplasm and then transported into chromatophores, where they assemble together with other, internally encoded proteins into working protein complexes that are part of the photosynthetic process.

Interestingly, the process by which these proteins are transported into chromatophores may also be novel and involve transit through an organelle called the Golgi apparatus, prior to becoming localized to the chromatophore. This suggests the occurrence of an initial, rudimentary process for proteins to cross the envelope membrane of the nascent chloroplast. This process ultimately evolved into one that is potentially more sophisticated and that uses specific protein complexes for efficient transport.

"This work demonstrates that P. chromatophora is a potentially powerful model for studying evolutionary processes by which organelles developed," Nowack said. "Obtaining a comprehensive list of proteins imported into chromatophores, including their functions and origins, as well as understanding the pathway by which these proteins are imported, could provide insight into the mechanism that eukaryotic cells use to 'enslave' bacteria and turn them into organelles such as chloroplasts and mitochondria."

International Development - Human Resources Business Partner

Role Summary

The HR Business Partner ensures that the client managers and staff in the assigned Cluster(s)/Unit(s) receive seamless, high quality, value adding HR services that support business operations while ensuring fair and consistent treatment of staff.

Key Responsibilities

The HR Business Partner will

  • Be the ?face of HR? to the Management Teams at Cluster and Unit levels in the assigned client areas effectively providing HR consulting, including bringing in other expertise and services from within and outside the HR function, as needed;
  • Understand the strategic business plans, issues and challenges of the clients, and their HR implications, develop client HR Strategies to support the business and to ensure timely HR solutions to meet anticipated challenges;
  • Facilitate and support excellence in human resources management in client areas by providing on-going HR advice to managers and staff on timely HR solutions, policies and practices objectively, with highest integrity, and based on best professional practices;
  • Apply best practices in human resources management to support performance improvements in the client units, including at team and individual levels;
  • Working closely with the Workforce Planning and Analytics Team, support and advise on development and implementation of effective workforce strategies and staffing plans, that are agile and relevant to meeting the business objectives of the client Clusters and Units; Use HR analytics effectively;
  • Support implementation of change and facilitate agility in the business units in a changing business environment using organizational development, change management and team building methods and techniques;
  • Train and coach client managers in good people management practices;
  • Advice staff on professional development and career management issues; and
  • Facilitate timely resolution of work place conflicts at the lowest possible level.

As a member of the larger HR Team, the HR Business Partner will:

Qualifications

Poll: Millionaire tax popular, spending cuts too

(AP) ? Most people like President Barack Obama's proposal to make millionaires pay a significant share of their incomes in taxes. Yet they'd still rather cut spending than boost taxes to balance the federal budget, an Associated Press-GfK poll shows, giving Republicans an edge over Democrats in their core ideological dispute over the nation's fiscal ills.

The survey suggests that while Obama's election-year tax plan targeting people making at least $1 million a year has won broad support, it has done little to shift people's basic views in the long-running partisan war over how best to tame budget deficits that lately have exceeded $1 trillion annually.

"Everybody should be called to sacrifice. They should be in the pot with the rest of us," Mike Whittles, 62, a Republican and retired police officer from Point Pleasant, N.J., said of his support for Obama's tax proposal for the wealthy. But Whittles said he still prefers cutting government spending over raising taxes because of federal waste and what he calls "too many rules, too many regulations."

Sixty-five percent of the people in the AP-GfK poll favor Obama's plan to require people making $1 million or more pay taxes equal to at least 30 percent of their income. Just 26 percent opposed Obama's idea.

Yet by 56 percent to 31 percent, more embraced cuts in government services than higher taxes as the best medicine for the budget, according to the survey, which was conducted Feb. 16 to 20. That response has changed only modestly since it was first asked in the AP-GfK poll last March. The question on Obama's tax on the rich was not asked previously.

The poll showed that overall, more people have a positive view of Democrats than Republicans, a ray of hope for Obama and his fellow Democrats with the approach of November's presidential and congressional elections. Fifty-four percent in the poll gave Democrats favorable ratings compared to 46 percent for Republicans, similar to results in January 2011, at the start of the newly elected Congress in which Republicans have run the House and Democrats wield a slender Senate majority.

Though embraced by congressional Democrats, Obama's proposal on taxing millionaires more has virtually no chance of passage by Congress in the political heat of this year's campaigns. But it stands as a rallying cry for Democrats ? about 9 in 10 of whom supported the plan in the poll ? and it contrasts with proposals by the remaining major GOP presidential candidates, who would lower the current 35 percent top income tax rate.

Obama has spent months touting his plan, nicknamed the Buffett rule after Warren Buffett, the billionaire who has complained that the rich don't pay enough taxes and that his own tax rate has been lower than his secretary's. The wealthy Mitt Romney, a leading GOP presidential contender, has released tax returns showing he paid a rate of around 15 percent the past two years.

Illustrating the wide acceptance for Obama's tax proposal for the rich, the poll showed it was supported by nearly two-thirds of independents and 4 in 10 Republicans. It also won backing from 6 in 10 whites and half of conservatives, two groups that traditionally are more likely to support the GOP, as well as by 6 in 10 people earning at least $100,000 a year.

Not everyone supports the idea.

"If their money goes to taxes, how will they afford more employees, better equipment, better vehicles?" said Republican Cheryl Mickler, 31, of Hope Mills, N.C.

As for the differing strategies for deficit reduction, more than three-fourths of Republicans and the largest share of independents preferred cutting government services. Democrats leaned toward tax increases, but by a narrower 49 percent to 38 percent.

Republicans have an 8 percentage point advantage over Democrats in the public's trust for handling budget deficits, essentially unchanged in recent months.

The GOP has the same edge for protecting the country, an issue it usually dominates. Peoples' trust in the two parties is about even for handling the economy, taxes and job creation.

Congress continues to receive dismal reviews from voters. Just 19 percent approve of the job Congress is doing, virtually unchanged from last December. That's not far from Congress' worst-ever approval rate in the brief history of the AP-GfK poll of 12 percent last August, shortly after Obama and lawmakers resolved a stubborn standoff over raising the debt limit.

"We put them there to do their job and they're not doing their job," said Gary Witalison, 54, a residential painter in Fish Creek, Wis. "They're not working things out. Work together."

The AP-GfK poll was conducted by GfK Roper Public Affairs and Corporate Communications and involved cell phone and landline interviews with 1,000 randomly chosen adults. It has a margin of sampling error of plus or minus 4.1 percentage points.

Stone Age pebble may be oldest engraving ever

A colorful pebble bearing a sequence of linear incisions may be the world's oldest engraving.

The object, which will be described in the April issue of the Journal of Archaeology, dates back about 100,000 years ago and could also be the world?s oldest known abstract art. It was recovered from Klasies River Cave in the Eastern Cape Province of South Africa.

?Associated human remains indicate that the engraved piece was certainly made by Homo sapiens,? co-author Riaan Rifkin of the University of Witwatersrand?s Institute for Human Evolution told Discovery News.

Rifkin and colleagues Francesco d?Errico and Renata Garcia Moreno performed extensive non-invasive analyses of the object. Methods like X-ray fluorescence and microscopic analysis enabled the researchers to examine every minute detail of the ochre pebble, which appears to have split off from a once larger piece.

The scientists conclude that humans intentionally made the sub-parallel linear incisions on the Middle Stone Age pebble.

?Upon engraving the piece with a sharp lithic implement, it is likely to have produced a markedly bright and dark red-maroon powder,? Rifkin said. ?The design may therefore have been strikingly visible shortly after it was produced.?

Ochre is a mineral-rich, naturally tinted clay that primarily consists of hydrated iron oxide. Ochre was among the earliest pigments used by humans and possibly other hominids for artistic purposes. Some even refer to it as the caveman's "crayon."

The Klasies River object measures close to 3 inches in length and contains a series of seven ?deep broad engraved lines and several, about 16 or so, narrower and somewhat shallower linear features,? Rifkin said. ?The fragment is a remnant of a formerly semi-circular ochre pebble that likely contained a much more extensive engraved design on its surface.?

Of particular interest now is whether or not the engraver made the design with symbolic intent. Use of symbols and meaningful images is thought to have been a significant breakthrough in human development. Language, math and countless other studies are tied to this basic skill, in addition to improved communication. To this day, art permits communication of identity and other things among diverse cultures.

Both linear and crosshatch engraved patterns may have been common thousands of years ago. Similar designs appear on engraved ochres from Blombos Cave, also in South Africa, and on ostrich eggshell fragments found in the Diepkloof Rock Shelter in the Western Cape Province. Some of these, and other, similar objects may even predate the Klasies River pebble, but studies on them are ongoing.

?The employment of red ochre for symbolic purposes likely played an important role in mediating increasingly complex social relations that emerged during the Middle Stone Age,? Rifkin explained.

Christopher Henshilwood, a researcher at the University of Witwatersrand, did not work on this study, but he has examined other very early probable engravings. For example, he studied abstract markings of another piece of ochre dating to around 70,000 years ago.

In that case, the engraving consisted of a more complex geometric pattern that looks like the letter ?X? repeated in a connected series.

The possible meaning of these lines remains a mystery, ?but they are symbols that I think could have been interpreted by those people as having meaning that would have been understood by others,? Henshilwood said.

At present, Rifkin and his team are studying 30,000-year old cave art from Africa. So far, they have determined that the abstract images depict a zebra, a rhino and a half human-half cat therianthorpe.

A great way to achieve better personal development is to respect yourself as well as others. If you do not respect yourself, you do not value the type of person you are and it will be much harder to improve your life. Respecting others is a great way to start respect yourself.

Personal development starts with a positive attitude. Having a positive attitude will affect every part of each day of your life. Everyone has their down days and troubles, but take a minute to step back, breathe, regroup, and tackle whatever situation with a calm mind and fresh outlook.

Take responsibility for your own life. If you aren?t where you want to be in life, it is easy to blame others. You may feel that your parents let you down, your teachers weren?t good enough, your boss had it in for you or that you didn?t have as many advantages as someone else. The truth is, your choices and actions have gotten you to the point where you are today. Gold coaxed into nanowires to allow inexpensive detection of poisonous industrial gases

ScienceDaily (Feb. 21, 2012) ? Researchers at the University of Pittsburgh have coaxed gold into nanowires as a way of creating an inexpensive material for detecting poisonous gases found in natural gas. Along with colleagues at the National Energy Technology Laboratory (NETL), Alexander Star, associate professor of chemistry in Pitt's Kenneth P. Dietrich School of Arts and Sciences and principal investigator of the research project, developed a self-assembly method that uses scaffolds (a structure used to hold up or support another material) to grow gold nanowires.

Their findings, titled "Welding of Gold Nanoparticles on Graphitic Templates for Chemical Sensing," were published online Jan. 22 in the Journal of the American Chemical Society.

"The most common methods to sense gases require bulky and expensive equipment," says Star. "Chip-based sensors that rely on nanomaterials for detection would be less expensive and more portable as workers could wear them to monitor poisonous gases, such as hydrogen sulfide."

Star and his research team determined gold nanomaterials would be ideal for detecting hydrogen sulfide owing to gold's high affinity for sulfur and unique physical properties of nanomaterials. They experimented with carbon nanotubes and graphene -- an atomic-scale chicken wire made of carbon atoms -- and used computer modeling, X-ray diffraction, and transmission electron microscopy to study the self-assembly process. They also tested the resulting materials' responses to hydrogen sulfide.

"To produce the gold nanowires, we suspended nanotubes in water with gold-containing chloroauric acid," says Star. "As we stirred and heated the mixture, the gold reduced and formed nanoparticles on the outer walls of the tubes. The result was a highly conductive jumble of gold nanowires and carbon nanotubes."

To test the nanowires' ability to detect hydrogen sulfide, Star and his colleagues cast a film of the composite material onto a chip patterned with gold electrodes. The team could detect gas at levels as low as 5ppb (parts per billion) -- a detection level comparable to that of existing sensing techniques. Additionally, they could detect the hydrogen sulfide in complex mixtures of gases simulating natural gas. Star says the group will now test the chips' detection limits using real samples from gas wells.

Also involved in the study were Dan Sorescu, research physicist at NETL, who performed computational modeling of the gold nanowire formation; Mengning Ding, a Pitt graduate student in chemistry, who performed experimental work and synthesized and characterized gold nanowires and measured their sensor response; and Gregg Kotchey, a fellow Pitt graduate student in chemistry, who synthesized some of the graphene templates used in this study.

Online coupons not always honored by merchants

When you buy an online coupon, how do you know a store will honor it?

Edgar Dworsky walked into Rose's Chinese Restaurant in Waltham, Mass., last week with a $6 coupon he?d purchased for $3 and found out the hard way that not everything is what it seems online.

The online coupon category is getting more crowded daily. Led by giants Groupon and Living Social, there are now hundreds of smaller competitors.?But not every email offering a discount is a good deal.

Dworsky purchased his coupon from MobileSpinach.com in early February. But when he went into Rose?s, the owner said he?d never heard of MobileSpinach and didn?t plan to honor it. Instead of enjoying a cheap meal, Dworsky found himself in the middle of the messy world of online merchant discounts.


Dworsky?s tale isn?t unique. Many store owners around the country say they?ve never agreed to accept coupons being offered for sale on MobileSpinach, and the firm has been dogged by nationwide complaints that it is selling allegedly ?fake? coupons.?

The firm?s co-owner, John Vitti, blames the complaints on misunderstandings, poor memories of merchants and over-eager affiliate salespeople, and says he?s happy to issue refunds.? But merchants getting pitched daily by ever-increasing number of Groupon-like sites are often caught in the middle, completely confused by the complicated world of e-coupons.

Dworsky was equally frustrated when he tried to use his coupon on Feb. 11.

?The man at the counter, the owner, said he didn't know what this certificate was, that he never agreed to offer these certificates, and that he had not been paid for them.?He indicated that someone had come in the day before with one like it also," Dworsky said.

And while MobileSpinach later agreed to refund the $3 he'd spent on the coupon, he wasn't really satisfied. Calls placed to other nearby restaurants unearthed a similar pattern.

"This is a scam of sorts ... or a naive company that thinks they can advertise deals that they have not yet formally acquired," said Dworsky, a former assistant attorney general in Massachusetts who now runs the consumer advocate website MousePrint.org.

Indeed, MobileSpinach has previously been accused of selling deals it didn't really have the right to sell. Last August, San Francisco-area foodie magazine Grubstreet wrote two stories about restaurants and consumers getting tripped up by Mobile Spinach group coupons that weren't authorized.?In November, a student newspaper at George Washington University reported the same problemin the Washington, D.C., area. The paper said 50 disappointed consumers were turned away from a small restaurant called Crepaway with invalid $10 vouchers they'd purchased from Mobile Spinach for $5.?

Other unusual stories dog Mobile Spinach. Jim Gilbride, who owns Old Country Deli in Hicksville, N.Y., told msnbc.com that a caller recently offered him an opportunity to buy a Mobile Spinach ad. He declined, and was surprised when the ad showed up on MobileSpinach.com anyway.

"I never heard him say (the ad would go up anyway)," Gilbride said. "I dismissed him when he called."

A man who answered the phone at Kabob Corner in Medford, Mass., said the same thing about a $3 for $6 coupon offered for that store.

"It is a fake coupon," he told msnbc.com before hanging up.

Even merchants who have dealt with Mobile Spinach seem to face some confusion.

At Creative Cakes in Silver Spring, Md., owner Randi Goldman said she agreed to a $5 for $10 in merchandise deal with Mobile Spinach about six months ago.? She generally feels pestered by sites like Groupon and Living Social, and doesn't like the revenue split they offer -- merchants only pocket 25 cents for every dollar in value that is sold.?But her Mobile Spinach salesman said she'd earn 100 cents on the dollar for every coupon sold.

"They said they'd pay me $5 for every coupon, and there was a special deal with the credit card companies who would pay the other $5," Goldman said.? So far, only a few coupons have been redeemed and her PayPal account has been credited the funds, she said.

Vitti, the co-founder of Mobile Spinach, admits that there have been some customer service issues, but blames them on confusion in the coupon marketplace.

"This space is getting crowded," he said. "There's just so much confusion. Sometimes merchants don't remember what they've agreed to."

That's his explanation for Dworsky's issue at the Chinese restaurant.

?I personally had a conversation with the owner of Rose's Chinese Restaurant and he apologizes for this confusion and so do we,? he said.?

A worker who answered the phone at Rose?s said the owner wasn?t present and declined to comment.

Vitti said the rash of San Francisco complaints was the result of a short-term experiment that involved offering deals for sale before they'd been arranged with a merchant. The company later ditched the idea, he said.?

He attributed the Washington, D.C.,-area complaints to rogue affiliates. Some deals on MobileSpinach.com weren't arranged directly by the Mobile Spinach sales team, but rather by affiliates in a revenue-sharing arrangement.

"There was confusion because the only way to redeem those was for people to make purchases online, and they were walking into the store with those," he said.?"Over time we were getting more and more customer support issues, to a point where we are uncomfortable with them. Sometimes these aggregators and deal providers don't have best relationships (with merchants). ... Sometimes you wondered, ?Who's got the relationship here?' "

So as of Feb. 4, he said, Mobile Spinach has stopped dealing with affiliates and will only promote deals sold directly by its sales force. The number of deals being offered dropped from "thousands to about 400" as a result, he said.

"We are trying to clean up this industry," he said. ?There has been many well documented problems in the coupon, gift card and now the deal space with redemption at any size merchant ... even large national retailers.? ?

He said the full value offer to Creative Cakes was real ? ?customer acquisition costs,? he said ? but added that it is only temporary.

Consumers who feel they've purchased a bad deal from Mobile Spinach should contact the firm for a refund, he said. "We have a refund-anytime, no-questions-asked policy," he said.

Merchants, however, face a slightly trickier proposition. They can refuse to honor any coupon, but that?risks irritating a potential customer. Gilbride, the deli owner, said he'd probably honor a coupon brought in by a frequent customer just to avoid a negative interaction.?

?I've been in this business 26 years now," he said. "I try not to get frustrated anymore by any of this."?
